The Colombia international, who is currently recovering from a long-term injury, will remain at Molineux until 2030, with his contract including a further 12-month club option.
The 23-year-old joined Wolves from Atletico Nacional four years ago as one of Colombia’s biggest prospects, but two bad injuries have curtailed his impact at the club to date.
Mosquera gained valuable experience during successful loan spells at both FC Cincinnati and Villarreal and was rewarded with his senior Colombia debut against Uruguay in October 2023.
Following an impressive pre-season in Marbella & Miami, Mosquera played every minute of Wolves' first five 2024/25 Premier League games under former boss Gary O'Neil, before suffering injuries to his MCL and ACL in a challenge at Aston Villa last September.
The defender's recovery has been positive to date, with a return to full training expected this summer.
Head coach Vitor Pereira said: “Yerson has the profile in a central defender that I like. He’s aggressive, strong, fast, good with the ball, and for me he has all the attributes to help us. I don’t have any doubts that he will fit my system.
“He’s injured, but every time he’s playing with teammates and giving energy. He goes to the stadium and tries to share his smile with everybody. This is the profile that I like to be in my side.
“The Mosquera that I see every day brings good energy to everybody. Inside the pitch is a player that can add something more to us. Outside of the pitch, he’s a leader with a smile, and I’m very happy for him and for us.”
