Emergency services were on hand at the junction of Ebstree Road and The Roughs to prevent vehicles entering the carriageway. A police car waited until the vehicle was towed away.
The car could clearly be seen on its side near the entrance to Dimmingsdale Garden Centre. The incident is believed to have happened around 10.30am.
Wolverhampton resident Ian Harris warned Penn residents on social media to steer clear of the area.
Posting a picture, Mr Harris said: "Road blocked by Dimmingsdale Garden Centre. Car on its side. Avoid if possible."

A spokesman for Dimmingsdale Garden Centre confirmed everything was back to normal by 1pm.
She said: "We are open for business as usual. Everything is fine."
